Preliminary Diagnosis of Malfunction in Liquid Chromatography Module

As is known to us all, the module malfunction of liquid chromatography instrument is very harmful, which not only wastes time, samples, energy and money, but also seriously interferes with the progress of laboratory work. Sometimes it greatly lowers the efficiency of experimental work, and even endangers the reliability of operation of laboratory. Therefore, when the whole module of liquid chromatography instrument fails, the troubleshooting is required to minimize the damage.

Here the preliminary diagnosis information of liquid chromatography module failure is summarized to help you to check efficiently and quickly.

Solvent

◇ Use brown solvent bottles to avoid mobile bacteria breeding.

◇ Replace the mobile phase in the solvent bottle of aqueous phase every 12 hours /24 hours to , and note the time.

◇ Use solvent of liquid chromatography grade as mobile phase and 0.45μm round membrane as solvent.

Open pump

◇ Check solvent bottles and solvent filter heads for contamination or blockage.

◇ Whether plunger rod cleaning function is open.

◇ Equilibrate the whole liquid chromatography system by an appropriate system and gradually increase the flow rate.

Close pump

◇ Flush all pipelines to remove possible buffer salts, sample residues or other solid particles.

◇ Flush system with proper LC preservation solvent.

◇ Turn off the pump after the pressure drops to 0 bar, then turn off the power supply.

Daily maintenance

◇ Change water phase every 0.5-1 days.

◇ Check if the plunger rod cleaning fluid needs to be replaced.

◇ Flush liquid chromatography systems using solvent matching applications.

Weekly maintenance

◇ Replace the solvent of the plunger rod cleaning bottle and check the mobile phase solvent and filter head.

◇ Check the back pressure state of LC system and replace the filter element of on-line solvent filter if necessary.

◇ Use a solvent matching the application method to flush the residue accumulated in the liquid chromatography system for one week.

Commonly used organic solvents

◇ If possible, maintain at least 5-10% aqueous phase in the mobile phase.

◇ Control temperature, avoid volatile organic solvents.

◇ Acetonitrile is easily polymerized under light for a long time to produce chain polymer, which will cause winding or blockage of each LC module , so it is necessary to ensure the replacement of new acetonitrile every 2-3 days.

◇ In order to avoid pump problems, please flush the instrument and pump with warm water regularly (>40℃).
