MCC Summary

Mixed-mode chromatography (MMC) is a high efficient chromatography which can optimize the design of the functional ligand structure, mix 2 or more than 2 interaction modes, enhance the target protein’s affinity and selectivity. Compared with traditional IEC and HIC, this new mode MMC has so many advantages like high-load, high selectivity, high efficiency and etc.

MMC’s characteristic

  • There are many interaction modes between MCC ligand and the target protein, such as the hydrophobic function, static electricity function, hydrogen bond function and etc.
  • MMC medium’s ligand has high density, and it has non-salt independent adsorption property which means people can adjust pH to elute the protein efficiently.
  • Protein can make hydrophobic group connected with MMC medium interacts with Ion exchange group: the static electricity interaction is the main function in low Ion environment, but the hydrophobic interaction becomes the main function in high Ion environment. While in middle situation, these 2 interactions are existing together.
  • There are 2 ways to elute the protein: when the hydrophilic combination is main, we can reduce the eluent pH by electrostatic repulsion; when the static combination is main, we can increase Ion to weaken the static interaction.
