What is Gel Purification Chromatography?

Gel purification chromatography is one of the main applications of gel permeation chromatography as a method of sample pretreatment.

Gel purification chromatography is a high performance liquid chromatography (HPLC) method which takes inert porous gel as the stationary phase, and it mainly separates substances according to the difference in molecular size based on the principle of size exclusion.

Separation Principle of Gel Purification Chromatography

As a sample solution containing various molecules flows slowly through a gel column, the molecules move in two different ways simultaneously: vertical movement and astatic movement.

Due to the large diameter, macromolecular substances are not easy to enter into the micropores of the gel particles and only can be distributed in particles. Therefore, the speed of downward movement is faster during elution.

Small molecules can not only spread in gaps between the gel particles, but also enter micropores of the gel particles, namely into the gel phase. During the process of moving downward, small molecules spread into the gaps of a gel, then enter into another gel. They keep spreading and entering constantly, so their speed of moving down is slower than large molecules. Large molecules in the sample flow out of column first, moderate molecules follow them and minimum molecules flow out finally.
